When upgrading the (-) negative side of my power system, should I make a connection between the alternator body and to the frame of the vehicle or should I just assume that the body of the alternator is grounding well against the engine and connect the engine to the frame. Also, if I make a connection to the frame, should I also connect that same point to the body of the car. Let's also say that I place a DMM at the point of connection at the frame and take a resistance reading at the point of where I will be grounding the amps at. If the resistance is low enough, will it even matter if I connect the frame ground to the body? I know I will be upgrading the Battery
(-) negative to the body but will I also have to connect that point to the frame also?

Yep
Try to hook your ground(s) to significant body parts Beware of the seatbelt bolts In some cars they are somewhat isolated from the body itself...
