I've got infinity reference 5-1/4 coaxials in the stock dash location in a 94 plymouth voyager. There are stock 6x9's in the rear hatch. I am going to hook up an alpine amp to the infinity speakers that does 60w x 2 @ 4 ohm, and the infinity set is rated at 45w rms and 135w peak. Do you think that the infinities will have decent midbass? And with the stock 6x9's in back, will there be some cancellation of midbass? I would just run the front but the rear fill helps a lot in the huge van. Do i need a bass blocker?

You should try to keep as much of your midbass up front as possible. I don't think 6x9's will cause much (if any) noticeable cancellation, but it will divide up your soundstage. Also, damping your doors helps your midbass a lot.
As for bass blockers, you shouldn't need them. The amp should have its own built in crossover anyway.
